La existencia de receptores se postuló para explicar el efecto de algunos fármacos en diferentes tejidos. Luego, se fueron presentando evidencias de que mensajeros extracelulares son reconocidos por moléculas de localización citoplasmática o integradas a la membrana celular para finalmente traducir la unión del ligando en la actividad celular.
